Could anybody with a kind soul just clarify which of these sentences is correct?

It would therefore be most helpful if you could write this essay in Danish.

Generally, sentence ONE.
Specific reason for sentence two:
If you have three or more helpful options then you could say “the most helpful” (with the unspoken “of the four options”).
But in 99.99% of cases it will be sentence ONE.
It could be argued that the word “most” is redundant as you only have a choice of helpful and unhelpful. But it IS a nice way of being polite.

I’m an English teacher of senior, advanced high school teacher and I guarantee that the first sentence is correct.
In fact, according to British English, the sentence would read:
It would, therefore, be most helpful if you could write this essay in Danish. (The reason is that the independent clause is ‘It would be most helpful if you could write this essay in Danish’ is truncated by the word ‘therefore’ which is being used, here, as an adverb, hence its positioning between two commas). I hope this helps.

Good catch – I missed that.
The “therefore” is a reference to a previous statement. Its inclusion is an optional factor in the sentence and, as such, needs the commas.
(As I just did with “as such”)
