Right click on Computer > Manage
Go to Diskmanagement.
See if there’s a disk without a drive letter, about 8 gigs in size. If it is there, right click it, and assign a drive letter.
If it ain’t, go for warrenty.

Then your USB stick is possibly broken. Try uploading a screenshot of the diskmanager.
TuneUp Utilities 2012 -> All Functions -> TuneUp System Control -> Administration -> Drives -> Advanced -> My Computer -> Settings -> Drive Icons -> Select All -> Ok -> Ok

Have you tried it on another PC ?
it probably has been formatted to a linux partition or has some kind of security feature enabled. non windows format is not recognized by windows. Boot to a linux live cd format the drive to fat32 or ntfs and the drive should be able to be used again
Linux mint live cd should take care of it
